Hey everyone, this Is my first post! Received my desire today from T-Mobile, and I was wondering if it is possible to wake up the phone via the bottom buttons instead of the power button. Previous phone was an iPhone 3g and I'm so used to having the button on the buttom to wake it up!

No, not yet. You have to hit the power button first.
This may come from a firmwear update, or more likely from a custom ROM after rooting the phone.

I am trying to find a way to turn off my HTC desire with only one press on the power button. Currently, after a long press on the power button, I have to choose shutdown and then confirm shutdown.

I'm not aware of any other way to do this, though it may everntually be part of a custom ROM.
Unlikely to ever happen unless rooted however.

I'm having an issue with my Desire, where I have to push down very hard on the volume - button for it to work properly. Volume + is fine though. Should I take my phone back to Telstra to have it fixed/replaced? I haven't dropped the phone at all, so I don't know why the volume down button isn't working properly.

Same here with volume +, had this problem after 2 months of use. Doesn't bother me much nor gotten worst since then.

I have just had the unfortunate wake up of my Desire power button not working, wonderful way to start the day.
I'm going to take it down to Telstra today and try and get it fixed under warranty, I don't believe that I have dropped it or let any water into it, but I'm worried that they'll say there is water damage and it won't be fixed under warranty (I found a few forums saying similar things)
What I'm hoping is an app or hack that will allow me to use another button as the power button. It is rooted (T-Mod) but I can't get into the bootup screen as I can't hold the power and volume button.
Any ideas or am I stuffed?

I couldn't find a way to power down the device via the hardware buttons, but there is a way to force a restart, after which you should be able to boot into fastboot/recovery and fix whatever issues you're facing. Subsequently, you should be able to shut it down from your ROM's power menu.
From the HTC website:
If HTC One X does not respond when you touch the screen or press the buttons, you can still restart it.
Press and hold the POWER button for at least 10 seconds, and then release it. HTC One X then restarts.

when you hold down the power button, you get three options. top one is power off, middle is airplane mode, bottom one is restart. the top one is the one you want and is the best your going to get as like you say, you cant remove the battery from these phones. dont see why you really need too either other than if the battery dies.
just hold the power button for 5-8 secs and a menu will appear.
Turn off fastboot in Settings. Press and hold the power button with volume down held until you get into the bootloader and select "power down"
Just another method of powering off completely.

After accident with water normally I have to asap turn off phone and remove battery. What to do with OneX? I would like to know solution before not after such case.

That is not possible. The quickest is via the menu apperaing when holding the powerbutton. You could download an app allowing you to power down the device via a shortcut on the homescreen.

I have HTC Desire with it normal firmware version "gingerbread". I am facing a problem that my phone power button does not work either when its display lights turned off, the power button is not responsive to wake it. I googled many forums and search many things but i did not find a solution for it. i put off my battery and then put on to it but my phone starts automatically and still power button is not responsive. please guide me ! or help me out.

I have same problem on my old Desire, now either get the power button fixed from service center or flash some custom rom/mod which enables track pad wake, I am using custom rom and can wake phone using track pad and its working just fine from last 2-3 months.

sounds like a hardware issue. If you have the tools, take the phone apart and make sure the lead is touching the button sensor.....It may have ripped or moved out of place I would guess.....
