If you have fastboot which is available in the apktool kit, you can verify bootloader status by putting your phone in fastboot mode (blue led) then open a shell and type "fastboot.exe getvar secure" without quotes. If it says no then you have unlocked bootloader. You can flash a kernel with flashtool or fastboot.exe. I recommend backing up your TA in flashtool prior to doing anything if you haven't already done so...could save you some trouble in the future.
When I connected my xplay in fastboot mode to PC and tried to flash kernel with fastboot, fastboot said <waiting for device>, can anyone help me???

There are a few things you must do
1. You must install flashmode and fastboot drivers
2.you must select xperia play as current device
3. Reboot into fastboot via ADB
4. Once booted into fastboot mode, click on select kernel to flash.

I know it's a pain to do but have you tried returning to stock and starting all over again?I take that flashmode works? That's what I would do if it happened on my phone:good:
EDIT Try disable antivirus
The real question is: have you run flashtool or the command line in "administrator mode"?

Check command which you are typing in cmd. Maybe you wrote something wrong. If you can paste your error after entering this command.

Personally I prefer to put the stock software (including recovery) on the phone prior to locking.
