Forgive me, I am new to "pure google" devices.
I am on build NRD90R. I used the nexus root toolkit to unlock the bootloader and root the device.
I am being prompted for a new OTA. Probably just the october security update. Do i need to "unroot" before i can take the OTA or will this overwrite what it needs to?

OTA will download, but it won't install if there are any changes to system partition. You could fastboot flash the factory image files if your bootloader is unlocked.

Noticed a new build was released for OTA. Doing a little research I discovered it won't work while rooted.
Question being, what's the easiest way to update? Temporarily run custom recovery and flash it? I don't want to loss root access or any data. If needed I'll re-root again, but rather not.

There are many ways to accomplish the same end goal.
You can actually install the OTA while rooted. Just need to run the small fix in this post:

Rerooting on this device is super simple.
There are also other options like flashing the factory images or one of the pre-rooted odexed/deodexed repackaged mostly stock ROMs. You can find them described here:

You need to run the fix I mentioned above OR unroot BEFORE installing the OTA or the OTA will fail near the end.

Don't quite understand the post, so better to just hold off on an attempt.. Would using the unroot option in SuperSU, allowing the OTA update to go through, then rerooting work fine without deleting data?
I'm know more about Nook Tablet than Nexus 7, didn't care what I did to my Nook because I could fix it. I'm more cautious with this.

Super-Su unroot never help you if you delete something in the /system directory
1)you must flash fix after root
2)do not flash permanent custom recovery and relock bootloader (if your bricked you devices you can send to Warranty Service)
3)replace backup of /system directory before update
and after update clean Dalvik Cache before flash Root again (if superuser privilegies still anavalable for you)
Understand the fix now. Noticed the update went through, but got an error when it finished. Flash ota update in temporary recovery. Correct?

I just downloaded the OTA to the device, unrooted, installed the OTA in TWRP, and then the latest superSU zip (still in recovery). Reboot and voila.

This is a Nexus device. There's almost no way to hard-brick it. Worst case, reboot into bootloader, and fastboot push the stock images.
The fix worked, and I'm still rooted. with all data. Temporarily booted into TWRP, flashed the fix and forced OTA update. Thank you!

LOL if you flash STOCK and then bootloader - you will get bootloop hardbrick without FASTBOOT and ADB !!! (only APX)

I'm not sure what you're saying in reference to my earlier comment...are you saying something I said was incorrect?
This is my first Nexus. If I unlock the bootloader and root, and stay with rooted stock, would a future OTA update relock the bootloader and undo root?
Also, are the OTA updates automatic, or can I choose not to install the update?

Once you are unlocked, you won't become locked unless you manually lock it. This is the case even if you accept an OTA or factory restore.
Some OTAs are mandatory like the one most people got when they turned on the unit, however most are optional and won't install unless you choose to, either on purpose or by accident.

However, to be fair, the DEB JLS36I update out there right now will cause you to lose root. It does not lock the bootloader, but it does whack root and custom recoveries. Unless you choose the option in TWRP to recover root after the install, in which case it rolls back the update and keeps both TWRP and root, meaning you are as you were as before. A few days later, you get the prompt to install the update again, and the circle of life continues.
Does that mean rooted and unlocked on JSS15R I can get the next OTA update but I loose root? I dont have any custom stuff like recovery or rom, just unlocked and rooted for a few apps like titanium backup, cpu Z and gemeni app manager

You can get the update and just re-root.
However if you have modified or removed any files, ota will very likely fail. Removing stock app would be an example of removing a file. On the other hand disabling stock app would be fine.
What if I just bought a nexus 7 LTE and already updated to to JLS36I? How would I get root then? Hypothetically of course...

Hi, my Nexus 5x has downloaded the new OTA but as I am rooted, bootloader unlocked and running TWRP I can't install it through the phone. Does anyone know which location this is downloaded to so that I can delete it? I can't find it anywhere. There is nothing in /cache.

You can download apk directly from chainfires site but you will have to manually update in the future.

I've got the OTA update waiting for me to select to get the November update but I don't want to risk losing the ability to unlock to bootloader if I ever want to in the future because I'm not sure of the bootloader unlockability for Bell bought devices. So if I unlock the bootloader do I lose anything, can I still get and take OTA updates from Google in the future?
You're phone will do a factory reset when unlocking the bootloader, so you'd want to back everything up. You can also still update with an unlocked bootloader.

No you can go to stock recovery and from your computer sideload the factory ota image of the updated software, its on the google dev site at the link below just go to the pixel xl section and the updates ota images are there.

Sideloaded last night. Didn't wipe anything. Will lose root but rooted phone right after update completed with no issues.
Should I install the image for Verizon or would it be more safe to use one of the other images?
I haven't been offered it yet, but I know it's coming so wanted to ask, trying to stay on the official channel.
If you accept the update will I lose root and have to re-root?
I'm out of practice since the Nexus 6P tether worked out of the box.

The OTA probably won't work because when you root with Magisk you flash a modified boot image. Process would be as follows before taking the OTA.
Flash stock boot image from the Image Version you are currently on (i.e. Flash the October Boot Image)
Reboot
Take OTA
Re-root using Magisk using the November Boot image to patch
add tethering line to build.prop or get the Magisk Module
Profit
You would need to repeat this for each monthly update to keep root.

Normally yes but I'm not sure if the partition structure is identical on the new Pixels but you could always try @topjohnwu 's method and report back.
I didn't think you could do an OTA with an unlocked bootloader, is that true? If so, topjohnwu's method wouldn't work anyway.
